Overview

PROVETOP provides integrated protein expression and production services for universities, research institutes, biopharmaceutical companies and diagnostic developers. Projects can begin with a sequence, accession number or target name.

We evaluate molecular weight, domain boundaries, transmembrane regions, signal peptides, disulfide bonds, post-translational modifications and downstream use before recommending a construct, host, tag, purification route and quality package.

Available routes include E. coli, mammalian and insect-cell expression, with other systems evaluated when required. Project records and milestone communication are included throughout the service.

Case Studies

Case 1: Mammalian expression of a complex glycoprotein

Project need

A human receptor extracellular domain for antibody screening and interaction studies, with multiple glycosylation sites and disulfide bonds.

Technical approach

Domain-boundary and secretion-construct design, mammalian transient expression, affinity purification and SEC polishing.

Project result

A high-purity, homogeneous target protein suitable for downstream antibody binding and molecular-interaction studies.

Case 2: Low-solubility antigen in E. coli

Project need

A low-yield antigen produced mainly as inclusion bodies, requiring a sample suitable for animal immunization.

Technical approach

Adjust expression boundaries and tag position, screen strain, induction temperature and time, then compare soluble-expression and refolding routes.

Project result

Improved soluble expression and a purified sample suitable for the agreed downstream immunization work.

Case 3: Cytokine expression with activity requirements

Project need

A recombinant cytokine with cellular activity for cell culture and in-vitro functional studies, with endotoxin control.

Technical approach

Secreted eukaryotic expression, multi-step purification, aggregation and endotoxin control, followed by a cell-based activity assay.

Project result

A sample meeting the agreed purity, endotoxin and biological-activity requirements for downstream cell experiments.
